You should get this in level 3 ("Level Zero"), because the training dummies you destroy in the level count as enemies. Since there are quite a few dummies in this level, combined with the amount of enemies you have defeated in the previous two levels you'll get this achievement in this level.
If you've skipped a few enemies/dummies, then expect to get this achievement around level 4/5, but it really shouldn't take any longer that that.

a really good level to rinse and repeat for this achievement is actually the first one: 'Po's dream' as it has lots of enemies but most are fairly weak.
