You will get the rocket launcher soon after fighting you first Griffon. You will be on a bridge and picking up a rocket launcher. Griffons will be flying at you left and right. When you can lock onto them fire away. Before going on I killed about 10-15 of them. Note that you dont have to kill youself as when I entered the next building at the end of the bridge the achievement unlocked.

This achievement is STORY BASED and must be completed to progress.
You are attacked by a Griffon when you make it to the top of the building you enter on your way through the stage. After you kill the first Griffon (on the ground as it is attacking you), you have to run across the open area to another enclosed area. In this next area you can pick up a couple of missile launchers. Step out of the enclosed area a short distance and press the
to enter Target Lock mode. When the missile launcher has a target lock you can fire the missile by pressing the 
and the rocket will chase the Griffon and hit it (assuming it did not hit the building or any towers). There is an additional stash of missiles on the Left of the next open area about half way to the other side. The missiles in this stash never run out so eventually you will shoot enough Griffons out of the air to unlock the achievement and progress in the stage.
Note: Don't forget to fill up that missile launcher before you go any further after the achievement unlocks - you will need them soon.
