You first need to unlock Professor Quirrell and use him to defeat the troll on the level Year 1 Chapter 2 - Out of the Dungeon.
You find the Professor Quirrell token in the pendulum room in Hogwarts Castle.
Once you have bought the character start the level in freeplay and use him to kill the troll at the end of the level for the achievement to pop.
