All of these will unlock by completing story levels except for Iron Man Mark 7, Agent Coulson, and Maria Hill:

Iron Man Mark 7 - will unlock once you complete House Party Protocol after the last story level.
Agent Coulson - will unlock once you complete Tabloid Tidy Up
Maria Hill - she will give you three missions on the Helicarrier deck that you have to complete to unlock her.

Edit: A comment has notified me that you may also have to unlock Taskmaster, however, this was not necessary for me and may be a cause of a glitch, either way, Taskmaster is unlocked in a location above the ocean west of the main island where the player can find a small box in a floating hovercraft in an altitude similar to that of the Helicarrier. He then can be purchased for 150,000 studs.

The majority of the Avengers characters will be unlocked as you progress through the story missions, you will have all of these by the time you finish story mode:

- Black Widow
- Captain America
- Hawkeye
- Hulk
- Iron Man MK1
- Iron Man MK6
- Iron Man MK42
- Nick Fury
- Thor

Once you finish story mode, multiple bonus missions will unlock. Completing "House Party Protocol" will give you Iron Man MK7, and completing "Tabloid Tidy Up" will give you Agent Coulson.

The final character to unlock is Maria Hill. On the outside of the helicarrier, you will have to find her and complete three mini missions. Her first location is in a tower on the northern side of the carrier, beat her in the race. The second location is under a slight awning slightly east from the HUB entrance, defeat all the enemies. Her final location is in another tower on the southern end of the carrier, beat her in another race.

All the characters unlocked outside of the story missions must be purchased for them to count.

Captain America - Unlocked upon completing Sand Central Station in Story Mode
Thor - Unlocked upon completing Bifrosty Reception in Story Mode
Iron Man MK1 - Unlocked upon completing Rebooted, Resuited in Story Mode
Iron Man MK6 - Unlocked upon completing Sand Central Station in Story Mode
Iron Man MK7 - Unlocked upon completing House Party Protocol(Bonus HUB Mission) in Story Mode
Iron Man MK42 - Unlocked upon completing Rebooted, Resuited in Story Mode
Hulk - Unlocked upon completing Sand Central Station in Story Mode
Black Widow - Unlocked upon completing Times Square Off in Story Mode
Hawkeye - Unlocked upon completing Times Square Off in Story Mode
Nick Fury - Unlocked upon completing Doctor in the House in Story Mode
Agent Coulson - Unlocked upon completing Tabloid Tidy Up(Bonus HUB Mission) in Story Mode
Maria Hill - Unlocked upon completing her three missions on the helicarrier.
