This has to be done in a single delivery.
Look at the market for an item with a demand over 60 (in case demand drops while you are manufacturing the item).
Make 50 items and sell as they are complete. If demand drops below 50 the achievement will not pop.
This is much easier when you have the perk that will show demand for the upcoming days. If you choose an item that you know you can manufacture within that time frame you should be able to sell with any issues.
