This is extremely important, if you played the game as the other characters and are doing a final run a cleric, most weapons that scale with faith can't equip runes. I played through this last, played through thinking I'd mostly be upgrading faith, with vitality and endurance as a minor. Like you'd normally think of playing a faith character.
You can do this, I did, but it makes your character weak IMO. You're better off just using this character magic as solace, and make a strength build instead.
Shelter is pretty good because it means you don't need health potions, but it takes so darn long to do that I couldn't use it on most boss fights cause it would be interrupted before I could finish.
Now if you get to the end and need to put the final rune in your weapon to kill the boss with a faith weapon like I did, you need to do the final proving grounds. I'll put a video of where it is.
The weapon is called Stare, it scales with faith, and it has a weapon slot for the ruin.
Also if you've played through the game already and just want to get through the game as fast as possible, check out this speed run of the game.
It shows a few glitches that no longer work so you'll actually have to fight the bosses he either skips or glitch kills, but the rest works fine.

The Achievement isn't buggy. What you have to do is using a two handed weapon(so no shortsword,dagger,fist weapon or every other one handed weapon)
I used the Scab (Staff) because it's really fast(120%) and the damage was over 70. It's not the strongest but with that speed you can do a lot of damage. It also has 2 socket's but I was a bit worried and only equipped the rune of Adyr.

+1 on the solution. Fortunately I kept a save just before the last boss fight in the Courtyard, and I just switched the Rune of Adyr from my short sword onto a hammer (NG+2 playthrough, was going path of the cleric even though I was using Agility-based armor and weapons). Achievement popped 5 seconds into the ending cinematic. :)

I just got this achievement without trouble on my NG+.
If you followed the walkthrough for this game on your first playthrough you'll have picked up an axe called 'Reminder' in a chest in the Chamber of Lies. I put the rune in this, glitched the boss, hacked away at him and it popped during the cutscene.

There are three endings in this game. Which ending do you get depends what you do with the rune of Adyr that YOU WILL GET when you continue the story. After getting the rune, talk with the blacksmith and then you can choose what you wanna do with the rune.
In this ending you need to put the rune in your weapon and make the killing blow with that weapon with the rune when you fight the final boss.
Remember that you need to put the rune in to your weapon the same way you put other runes in them. Talking with the blacksmith about putting it to your weapon doesn't put it to your weapon. You need to put it yourself manually.

This trophy is bugged on dual wielded weapons. Put your rune in a one handed weapon and redo the fight if you dont get the trophy you expect.

***SPOILER ALERT***
Near the end of the game after fighting Annihilator you'll be given a rune in the Palm of God. In order to get this trophy u will need to put Adyr's Rune in a weapon slot. You don't have to fight entire battle with that battle. What I did, is hit the boss with my high DPS weapon and did my killing blow with another weapon which had this rune on it.

This trophy is glitched for me, putting Adyr's rune in my weapon awarded me "God is dead" instead.

This is for anyone still playing this game 9 years later having trouble with the final boss, especially on NG+ (That was crazy tough. I think my Health was too low, and no way to respec. And at 250K+ per level I wasn't gonna farm the points lol.)
All the other guides info applies, but here is a Glitch that as of October 12th 2023 at 1045Pm still works.
