Fortunately for you, this trophy doesn't refer to just the "ghost" enemy in the labyrinth, but all ghostly enemies in general, so the skeletons count too. You should get this through natural progression.
If you have not played the end game area of the base game you won't recognize them but the skeletons are a re-skin of the citadel soldiers, and therefore pack massive damage for faith and agility builds. Watch out!

You can access the Ancient Labyrinth DLC by talking to Crafter the after defeating The Beast.
The Labyrinth is filled with transparent ghosts - for which you need a magic damage weapon, like The Heirloom - and glowing skeletons, which you can damage with 'normal' weapons. They all count towards the achievements, meaning you'll likely get this just by hacking your way through the DLC.

I was counting ghosts and skeletons as I killed them and the achievement popped after 21 kills..lol
