You will start with some cards, but you will likely need more from either packs or the Auction House. The best way to do this is to look at your Item Binder, and pick Team or Chemistry from the left side. This will pull up a list of each of the teams/chemistries and the number of each that you have. Then you can go to the Auction House and select a player for one that you have 2 in.
If you're looking to upgrade your team on the cheap, go to "More Packs." You'll notice that there are Silver Player Packs for 3200 coins, and Get A Player for 500 coins. Some quick tips:
-Silver Player Packs give you at least 1 65+ silver+, and 3 silver+ players.
-Get A Player gives you 1 silver+ player
-You can buy 6 Get A Player packs for less than 1 Silver Player Pack.
-For the mathematically challenged, that means you get 2 extra players.
-In the six packs I bought, I never got less than 65, and pulled a 73 gold.
As soon as you put the players in your lineup, this will pop.

In order to activate chemistry, will need to have the minimum number of players with the same chemistry in your starting lineup plugged in at their natural position. The minimum number of players various by chemistry. Most player cards have a chemistry associated with it and it is displayed on the front of the card on the right side under their rating.
For example, once you have 3 players with your favourite team's chemistry in your starting lineup you will see the chemistry activate on the right side of the lineup screen. In order to unlock this trophy, you will need two chemistries activated at once which is fairly simple to do.
If you completed Longshot and bought a single player from your favourite team (the team you chose during the MUT tutorial which automatically gives you 2 players from that team) then you should already have enough players to activate the Longshot chemistry (both Longshot players) and your team chemistry (3 players from the same team). In MUT mode, your players will have formations they are best suited for (long pass, run stopping, etc...) and you just need to collect enough players suited for TWO different styles of play. Then go to lineup assistant and create a new lineup based on chemistry (one offense, one defense). You can collect them naturally or just buy them from auction.
