I changed the sliders to Rookie to help me get this.
Confirmed modes you can get strikeouts to count: MLB Today, Play Ball, Franchise, Postseason
Confirmed modes you CAN'T: Online, Online Leagues, My Player Mode.
Basically you have to get 200 strikeouts, and theses will accumulate and come easy over time.
To track these you can go to the Main Menu > Features > V.I.P Viewer and it will be under the pitching tab.

Just to clarify, ranked matches do not count as well, correct?
