You must have upgraded the assassination techniques marked with a * first!
Play through the level until you get to the locked door.
There will be two guards patrolling the ground floor with more on upper levels.
Ground floor:
1. Dangle from a pole and perform a Hangman's Hymn* assassination.
2. Wait until you terrify the second enemy with the hanging body so you can drop on him from the ceiling and perform a Bat's Prey* assassination.
First floor:
3. While looking out of the vent drag the third enemy in and to perform a Prowling Spider assassination.
4. The forth enemy does not move but is looking around. Perform a standard Silant Assasin assassination.
Second floor:
5. There are two enemies. Destroy the left lamp above the enemies so you can get on the same floor and hide in one of the hiding spots until a enemy passes so you can kill him from the hiding spot and perform a Brief Shadow* assassination.
The achievement unlocks after the fifth kill.
Shiftie
You will need to get a few new techniques in order to get this achievement, though you can get this around level 4ish so it's not much of a wait. What you need to do is just get the first 4 techniques (Hangman's Hymn, Emperor's Abyss, Twilight Gate, Bat's Prey) and use each of them, as well as you're regular stealth kill. So, kill someone with a normal stealth kill, kill someone from the opposite side of a door, kill someone while you're hanging under a walkway that they are on, kill someone while you're hanging from your grappling hook and jump down on top of someone while you're hanging from anything. Leap of Faith also counts as a unique kill.
