Comes naturally through the game as you go down in the caverns to find the guns. You don't even need to kill the moles, your party killing them counts too.

In the mission "First Step In Prison" you will access the Drilling Well where you must eliminate some moles, you will most likely unlock the trophy here if you find and kill them all. Moles appear many times during the playthrough and you may want to try and sneak kill them (Assassins skill) since they are tough opponents if you fight them head on.

Story related, unmissable.
You will get all 15 kills in the Drilling Well area of the Crater while doing the Weapons for the Battle story mission. They aren't overly difficult, aside from attacking you and your two allies in rather large groups, but there are a few explosives that you can loot off of corpses in the caves, and you will also have access to your first ranged weapon - the nailgun. The moles come in several different sizes, with the larger ones obviously being the most difficult, but you can start each fight off by using stealth mode () to open the battle with a powerful attack on the first enemy.

In total, there are about 1000 enemies in the story of the game, you kill everyone (almost everywhere the main condition for moving further in the level), one might say, is a storyline.

In the first chapter, when you get into the caves, there will be enough moles there to get this achievement.
But even on the simplest difficulty, EASY, if there are more than two enemies, then problems arise with eliminating them. So be prepared to start the game from the last save point many times.
