Enimies are pretty easy, so I would suggest you get a feel for each characters ability and rank up those you think are best. You can even replay missions to gain XP and level up a bit to make future missions easier.
I personally used the load out of Iron Man, Wolverine/Juggernaut (after unlocking him after chapter 2), and Thor Goddess of Thunder (which later I switched to a better load out, but for this chapter, she is best).

Story Related. Cannot be missed.
You will get this after you defeat 'Rags' in the last fight of the last mission called "Dark Avengers", which is in the chapter "H.A.M.M.E.R".

Story related and cannot be missed. Complete all the battles in all the five stages of the H.A.M.M.E.R. chapter of story mode - H.A.M.M.E.R. (11 battles), Juggernaut (eight), Venom (eight), Oscorp (12), and Dark Avengers (10).
As with all chapters, beating a fight will unlock the next battle(s) and beating all the battles in the stage will unlock the next one, and once all stages are done the next chapter will be available. See the roadmap for hints for getting through battles easily.
