1) Some of the missions won't become available until after you complete certain story related missions first.
2) Do as many of the side missions as possible! If you don't you may be cutting it very close by the end of the game or not get your achievement at all.
3) DLC does count toward this achievement.

*QUICKEST SOLUTION:
1) Complete the first set of Citadel assignments.
2a) If you want Liara as your squadmate then complete the "Find Liara T'Soni" mission.
2b) Otherwise complete "Feros: Geth Attack" and all of the Feros assignments.
3) Complete remaining Citadel assignments.
4a) Begin completing UNC assignments in the order you received them until the achievement pops.
4b) Feel free to continue the main missions as well but STOP after the Virmire: Assault mission before you travel to Ilos as it is the point of no return and you will not be able to complete any secondary assignments after it.
Depending on which squadmates you choose, you will also be working on the following achievements:

I highly recommend doing every available side quest at the citadel near the start of the game. Not only does this help a lot towards the completionist and ally achievements, but it's a great way to boost up your XP levels before heading out into the galaxy! Also, many of them can be done in a matter of minutes so you can blast through them all in 2-3 hours.

Thanks Rich! I believe about half of the Citadel assignments cannot be started/completed until AFTER you have completed one of the main missions. After doing one of the missions, however, I think they all become available.

Wouldnt it make sense that if you unlock the ally achievements for completing the majority of the game with THEM, that you would also unlock the Completionist achievement at the same time? Bc that def. did not happen. Do they consider a majority of the game for Shepard to be more than the majority of the game for an ally?

There must be different progress tracking for each character. I unlocked the Kaidan Alenko Sentinel achievement first, then 1 mission and 5 assignments later I unlocked the Completionist achievement; and I have to still unlock the Garrus ally 'chevy for this play through. I'm currently done with all available side missions and I'm on Ilos so I'm hoping to get his before I finish the game and I have only played with them in my party.

I was trying to complete as many of the side quests as possible, and unlocked this by the time I finished both Noveria and Feros. I hadn't yet gathered enough to finish the "collectibles" missions, but had only a handful of outstanding side quests open at the time.

After multiple playthroughs I finally managed to do best and fastest possible way to acquire this and the others majority achievements. I played this on casual and skipped every possible conversations and after I finally unlocked majority achievements turian and quarian ally there was little less than 8 hours in game clock so with this list you should get this fairly quickly. It should be also noted that my Shepard was earthborn but that should only be linked to few missions and every ''Shepard'' has his own missions that goes towards this achievement.

It seems that if you are going towards squad member achievements the main thing is to END the missions with squad members. With this said all the missions that will end talking to Hackett in Normandy are useless.
Another thing that should be mentioned that if you do Wrex or any other squad assignments take that particular member with you because that way the mission will end as soon as you get your goal.
EDIT. There should also be noted that you can end some assignments just ending them right away. At least scan the keepers and that mission where you have to get bhatias wife's body you can end at the start.

It seems that if you are going towards squad member achievements the main thing is to END the missions with squad members. With this said all the missions that will end talking to Hackett in Normandy are useless.
Another thing that should be mentioned that if you do Wrex or any other squad assignments take that particular member with you because that way the mission will end as soon as you get your goal.
EDIT. There should also be noted that you can end some assignments just ending them right away. At least scan the keepers and that mission where you have to get bhatias wife's body you can end at the start.

To unlock this trophy you need to have completed at least 45 assignments and pretty much all of the main story missions. Depending on which squad members you have on your party while completing your missions and assignments, depends on which 'Ally' trophies you will unlock.
If you want the 'Asari Ally' and the 'Turian Ally' trophies for example, follow their guide while only ever using Garrus and Liara on your team once you have recruited them. Liara will take longer than the other members as she requires 50 assignments.
Do not try to do this for Ashley and Kaidan together in one playthrough, you won't complete enough assignments for them both, so use them in separate playthroughs.
You will have to follow their guide for a total of three playthroughs to unlock all of the 'Ally' trophies.

By majority of the game, they mean an undefined amount of the game. I recommend that you choose two characters and go with them till the end. Try to not start a side quest without the characters you want. There around 70 side quests including the main quest ones. So, make sure you do around 60 of them before Ilos. IF you haven't gotten the achievement after that, then you should try exploring the galaxy and doing more quests. Remember to talk with allies in order to generate more side quests.
NOTE! Completing the DLC makes a noticeable contribution toward this achievement. You should be able to earn all of the ally achievements before you get to Ilos if you complete the DLC.
