Quests are optional objectives that are tracked in Roguelite. There are three objectives active at a time, one in each of three categories:
-Play as ___ character ___ number of times
-Win ___ number of rounds total (individual waves of a stage)
-Win ___ number of stages (must win the final wave)
Win 100 for this achievement. As of now, there is no way to get a new quest objective except completing it. If you want this as fast as possible, you can start a level with the quest character and immediately quit. Rinse and repeat.
Edit: others have noted that their quest for character has not been pulling from their pool of unlocked characters. Since there is no way to get a new objective, I would focus on going for Leader blocks exclusively to increase your chances of unlocking characters.

There are a lot of hero's in this game. I think maybe the first one is completely random or maybe it can select from your pool of DLC hero's even though you do not have them unlocked yet. Because after I got the King I did not get another quest for a hero that I had not unlocked. So it probably is because of the DLC.

My starting quest was play as king 1 time. My starting leader unit was the new Crusader class. I've unlocked 6 other classes, none of them have been King. I feel robbed that you didn't go through the same struggle as me lol.
Jokes aside, figured I'd post and say the starting quest for me was not from my pool of classes.
