Typically, this will wait until you've unlocked the Stygian Guard and have access to Spell Weakness and other powerful spells. Either one stack of Spell Weakness and a spell like Crpyt Builder dealing at least 60 damage naturally, or multiple stacks to lower the requirements.

However, there are a few other ways to get this early, such as the Hellhorned Battering Ram, where it would need to be used on a floor with at least 30 armor across all units, or a powered-up Awoken Restoration Detonation healing for 30 damage.

I had a spell that did 100 damage to everyone in a row and I upgraded it for a +20 magic bonus with the hellbender cards.

second hellhound champion imp queen with summon imps and imp spell (+10 damager per imp) is really easy too

The spell he’s the other commenter is speaking about is a Hellhorned spell called Inferno, it deals 100 Piercing damage to all units in a floor (included yours), he’s right that you can add +20 spell damage (or maybe Double Stack, never checked) but as it costs 3 you might end up having to end your run by killing yourself to the point you can’t recover by having to use it when it comes up during an inconvenient draw. It’s the easiest early way to get this, but the solution isn’t wrong that with the Stygian Guard you’ll often get spells doing 200+ damage, they’ll likely be your first faction unlock so there’s no need to rush. The main problem with Stygian Guard is that you’ll probably lose a lot early on learning how to use them. You’ll still unlock this even while losing though.
