By far the easiest method to getting this achievement is with the following:
-2 controllers
-Danger Kustom Kombat modifier
Provided is a quick video solution and a text solution.
Text Solution
Play as Erron Black (any variation works fine) and using your second controller fight a dummy in Kustom Kombat. You do not need a second profile, you can log in a second controller and select the option to 'continue without saving'.
With the Danger kustom kombat modifier, it lowers all kombatants health to 1%, allowing you to simply throw out caltrops and get the achievement. This can be done even faster if you go into the gameplay options and set the Rounds to Win to 1 round, otherwise you'll have to throw 2 caltrops.
To get the Danger modifier, you must have completed the Hourly Living Tower that plays with the Danger modifier. Unfortunately the Towers are generated randomly every hour so there is no way to log in a certain time and see the Tower that will get you this modifier other than to check back hourly and hope you get it.
If you don't have the Danger modifier
Simply continue as normal with two controllers with the Rounds to Win set to 1 round in a two player 1v1. Beat up your dummy account with Erron Black and then throw out the caltrops under their feet and wait for them to die.
If you don't have a second controller
Load up a Single Fight in the Single Player menu and choose Erron Black, and any opponent on Very Easy difficulty. I usually choose Sun God Kotal Kahn as my opponent as he doesn't have pesky projectiles. Beat up the opponent until their health is very low and then throw the caltrops. Try to get the CPU opponent to stand on them as much as possible during this time and pray they don't move very far.
If you can enhance the caltrops they'll do even more damage and ensure you'll get it against the CPU.

For this achievement, I suggest you start a Single Fight on very easy. You are going to play as Erron Black. His caltrops ability is caused by

depending when you are standing, the left/right is always opposite to the side your enemy is. Simply beat your enemy down low and spam caltrops hoping he/she will step on it.
If you have 2 controllers, you can also do it that way

Is it possible to have 2 controllers and 2 profiles and just do it that way?

I have no idea, I don't have 2nd controller, but it seems like it. Achievement isn't game mode specific.

Caltrops? What the heck are Caltrops? Never fear, this is pretty easy. And when did this game learn what a 'C' was? Kaltrops? Anybody?
By far the easiest way is with two kontrollers, being Erron Black (any variation) vs Anybody.
Beat up P2 and then when they are at low health, get klose to mid distance away and throw the caltrops (spiked metal devices thrown on the ground primarily to impede horses or wheeled vehicles) by pressing
if you are on the left of P2, or 
if you are on the right.
Erron will throw caltrops on the floor, killing P2 and getting you the trophy.
If you want to get it quicker and have unlocked the Danger Modifier, play Kustom Kombat and aktivate it, both players start with 1% health. Simply input the former button sequence and it will kill P2 immediately.
You kan get it vs the CPU, but it's much easier with two kontrollers.
