As of 7 PM PST on January 2nd, 2020, this achievement is patched.
It can be done in a private lobby with as little two people, each loading up three controllers on the main screen (press Y on the main menu after you sign in guest accounts). It can be done with three people with two controllers each, or whatever combination you want to have 6 "human" players.
From the main menu, once you have your guest accounts loaded in, you'll want to select the following:
• HOST CUSTOM GAME
• HOST PRIVATE LOBBY
• Infection (Miners vs Player Zombies)
As it is a private lobby, you will need to send game invites to any friends you are boosting this with. Once they and their guest accounts are loaded into the lobby, you can press START.
Once in the game itself, as long as your main gamertag (not the [1] or [2] guest accounts) is not a zombie, you'll be able to unlock the achievement. Have your dummy controllers and anyone who is not getting the achievement to get together, then have the player in control of the zombie infect them all. It should pop immediately once you are the only "human" survivor.

Have you tried buying the DLC that costs more than the game? LUL

Shout out to the developer for releasing patches so quick. It’s rare to see that. Hopefully other developers follow suit
