This trophy is cumlative and can be done over multiple plays.

In order to get the clown pill, you must first find the clown (See Six six six for map) and then get him to a helicopter spot without him dying. When he is safe and boards the helicopter, he'll give you the the pill.

Once you have it, you are able to activate it by pressing + , causing you to swallow the pill and turn into an evil clown. While in clown mode, you can only use hand attacks, but they are more powerful and you move much faster. You can not pick up any weapons or special survivor coins while in this mode. It will wear off after about 15 seconds, so make sure you activate it when there are plenty of enemies nearby to maximize your kill output while in that form. Rinse and repeat until you have 300 kills while in clown form and the trophy will pop after the final one.

You can see how you are progressing by going to the trophy room and going to the spot where that trophy will be (8th from the right).
