The first and only time (so far?) I got a diamond was from a boss in "The Mines in Eufaula Desert" (aka Ingall's Mine). It was part of a mission where you had to rescue some people from a collapse.
I recommend using the hammer Remington gives you near the beginning of this quest. Apart from its insane damage (at the time) it has a knock up/back effect which essentially stunlocks everything including bosses.

I was picking up animal feces in the cow pens and found 1 randomly.

So I picked up two diamonds from picking up poop in the cow fields randomly lol
