Sasuke's secret room can be found in the lake area in Konoha (home village). You will not be able to access it until you have the walk on water ability.
Go to the dock area where Sasuke usually stands, and jump across to the middle island. Turn right and you should be able to jump down onto a small beach (not the one you jump on to get onto the island). From this beach, you can walk on water turning left (facing from the island) to see a cave. Inside this cave you will find his room.
