Most of the Werewolves reside in the Vellosk. I got the achievement at about level 42.
Kill 500 of them. I went round and round some of the alter spaces and just killed them 3 at a time. It took a while but then it popped.
Sure there are werewolves in the other levels but not in sufficient quantities to grind it out in a short space of time.

Although the achievement states you must kill 500 Lycanthropes, and normally this term is used synonymously with werewolves, in this game it also applies to the rat people. So most of the levels where you go into the sewers, mainly in Black Lake {Mission the Dead Rats}, there are plenty of rat people, and at a much earlier level then Vellosk which requires you to be at 37.

You'll get this trophy for killing 500 enemies classified as Lycanthropes. Lycanthropes come in a couple of names, the earliest being the Wererats.
The best places to find Lycanthropes are the Vellosk and Blacklake District adventure zones. They make up most of the enemies in Vellosk, but you can also kill Wererats in the Blacklake District (across the bridge by the campfire where the alchemist is and left of the Waterfront area).
To check your progress on this trophy first press the Options button > Quests > Journal > press R1 to go to Achievements > Slayer > hit
to show them if they aren't visible and scroll down with R-stick to the Lycanthrope Slayer entries.

When facing the 'Wererats' up to and including escaping the Spellplague explosion all will count, but these are a limited resource used in one-time only adventure zones. The best way to achieve this trophy is kill the humanoid 'Werewolf' tagged enemies in Vellosk. One of the best circuits to run is a circle around the 'Elder Rise' area of the Vellosk map.
Extensive testing has shown that the wolves marked 'Werewolf' do not advance your kill counter.
