This is pretty simple. When putting together your lineup, you'll see the little gold synergy symbols on the bottom of each card, and along the bottom of your screen you'll see each of the logos with a fraction next to it (ex. 2/5) This means you currently have 2 people on your lineup with that synergy, but you need 5 to activate it, and give that particular stat boost to those players.

To get the achievement you just need to edit your lineup, make sure at least one is active (which is just about guarenteed. You'd almost have to try not to), save the lines, and the achievement will pop.
