Mission 1
When you first encounter the Spider and your master throws the Dual blades towards you, and Ken knocks the Spider to another building rooftop.
When you are about to use your Grapple for the first time, the bats will appear to attack you.
Use your R Trigger+Y, Ken will elevate up and spinning the blades. This will instantly grant you 24 kills. Quickly use your wind Ninjistu to kill the remaining you missed, this should grant you 30 kills.
Immediately, when you land on the ground, use your Ninja Vision to detect the big flying bat monsters, and use your elevating attack again to continue your combo. While on air use your wind ninjistu again. This will surely grant you 50 kills
Basically you just have to kill before the kill hits runs out.

Thanks for the solution. Once I got on the 1st platform after the QTE where the bats came I just waited till the bit me a couple times to give others the time to come my way. Then I did the R Trigger +Y. After that I did the Ninjistu spiners, then a Ninja Vision to get the remaining all on the same platform. Thumbs up from me anyways. ;)

In Mission 1:
Just after you are given the dual blades, you will be on a rooftop with some bats attacking you. Destroy these, and there will be a second wave of them along with some bird enemies. Destroy them as well and grapple to the next rooftop. Continue on and grapple to the next rooftop as well.
It is on this rooftop where you will earn your achievement. There will be a group of bats nearby and another in the distance. Make sure you have a full energy bar for ninjutsu. Simply run into the first group of bats, tap B to activate your wind ninjutsu and press LB to activate your ninja vision as your ninjutsu is killing the bats. Now, under the power of ninja vision, RT run to the second group of bats and activate your wind ninjutsu again. You will end with about 60 kills and your achivement.

In the first mission, chase the spider type carrier after you get the falcon knives. Grapple hook to the next building, run up the wall and you'll see two groups of bats. Approach the one near the spider slowly. They will start flying towards you. Now head backwards to the first group. Wait until most of the 60 Bats have surrounded you and use Cyclone shuriken to kill them in an instant.
