After gaining access to this new form during the challenge rooms in the DLC arena area, you have to work on the form-specific missions to rank it up to "S".
Unfortunately, the arena challenges in the DLC do not count towards form-specific missions so you will have to go out into the base game content to achieve these objectives.
My coop buddy and I had great success with the form-specific missions farming the demi-dungeon "Dark Tower".

The Killer Bee is unlocked by obtaining 14 total medals and then going through the second door in the Colosseum. Once you unlock it, you'll need to (fast) travel back to the main world to be able to work on challenges (Form challenges are locked inside the Colosseum). Just like in the base game, you'll have a series of challenges to complete while making use of new attacks and passives, and completing them all is required to reach S-Rank.
Thankfully, just like with the base game, the northeast corner of the map is a great place to grind out challenges. In fact, this Form's challenges are all very easy and straightforward, and all can be done in that spot (refer to the image in Job's done! (25G) or Super Wiz (100G) if you are unsure where I mean).
In fact, because all of the Killer Bee challenges are so easy and straightforward, I won't bother detailing them all. Instead, here are a few I wanted to add a small amount of detail or tips on.

- Killer Sting III: Kill baddies with Killer Sting while you have 3+ Attack Up buffs - rather than relying on honey drops from enemies, simply equip Pump Up (and Magic Life if you want), and as soon as you spawn in the area mentioned above, use 4-5 Pump Ups so you can then go to town on enemies without worrying about it dropping below 3 buffs
- Custom II: Killer Queen: Hit baddies with Familiars - equip Magic Hat and just spam Familiars in the area mentioned above
- Custom III: No Bee Like Me: Status baddies with Hive Swarm - if using the area mentioned above, drop the Hive Swam early and in the center, trying to avoid the drop of the hive from hitting enemies so they don't die before a status builds up. You should equip all three of these passives: Poison Tipped to poison them, Stun Powder to stun, and Spooky to build fear. This should inflict all three statuses on each enemy, making the challenge go by much quicker
