Not that this really requires a solution, but I figure I would explain the achievement requirement since the achievement descriptions in this game are very cryptic.
This achievement is unlocked for reaching a combo multiplier of 50.
While you may get this naturally, there is an easy way to boost your combo meter during the first boss.
During one of the boss' attack patterns, it will release 4 bugs from it's mouth. You can quickly kill these and then hit the boss periodically during its bounce phase to keep your combo going. If you can keep this up long enough (and use the default weapon to limit damage), you can easily exceed a combo of 50.
You need to kill an enemy to increase your combo number, but you can prevent your combo meter from depleting by simply attacking an enemy.
