This one has to be done in level 12, "The Sanctum." You are unlikely to get it in one playthrough, although if you are going for all the achievements you'll need more than one anyway.
If you do want to grind it out, it counts if you scare off the same ones repeatedly. You can find a group and shine the light on them to scare them off, put it away (let go of the right stick) and then point it at them when they return over and over again until you hit 500.
