YOU CAN GET THIS IN THE 1ST LEVEL

If you have played a Dynasty Warriors game or anything like that, then you know a 1000 enemies is no big ask. Juts use X and Y and do some easy combos and you will smash through the enemies in your way with ease. In the 1st level I got 10,000 enemy kills without really trying, so you will get it easily as well.

You will most likely earn this through the tutorial fight where you and Kid are running through Wano Island looking for Kaido. The amount of enemies that you can find throughout the game makes it to where you will come up with this multiple times. Furthermore, you will need an enemy slain rank of 1500+ for most S rank episode requirements, so it doesn't hurt to just kill as many as you find at times.

You will likely earn this naturally. Certain episodes feature massive amounts of enemies while also featuring boss fights. So you will likely end up defeating 1000 enemies while just trying to defeat a boss character. Either way, simply clear out 1000 enemies in a single episode to earn this.
