This is a cumulative record that can be found in the Results tab of the pre-battle screen. The first page will show you the total amount of enemies you've slain, and seeing as how this will be near the end, you'll have to topple plenty to get the Platinum. Wait until after you've obtained the Rich Pirates trophy to see if you still need to work on this, though I highly doubt it by that point.

You will definitely achieve this if you are going for the Platinum. Warriors/Musou games are all about clearing large amounts of enemies with attacks. Simply play the game and you will earn this naturally. I earned this after the Dramatic Log while going through the Treasure Log. If you are curious, your enemies defeated counter can be viewed on any Battle preparation screen by going to Player Information and selecting Results.
