Getting kills is the point of the game, so this will come naturally from traps, spells, guardians, and personal (bow/sword) kills. When I finished the main game, I had around 9,000 kills, and I hit 17,000 after getting all skulls on War Mage. You will likely either need to farm levels or try to finish Nightmare to get this.
Note that guardian kills (even those already present in some of the levels at the start) DO count. Getting kills and then losing a level DOES add to your total. Unfortunately I'm not sure about getting kills and then re-starting a level.
UPDATE: When I finished Nightmare mode I had 29,927 kills. I replayed one earlier level and then got this final achievement! It will pop in the middle of a level, but I don't think the leaderboards will update if you quit the mission before completing or failing it.
