Simply bring the Vertex SMG along on a heist with some medic or ammo bags to help you survive and simply gun down officer after officer. Try to keep count of your kills as I believe the achievement unlocks when you go to escape, not when you received your 100th kill.

The Kross Vertex Submachine Gun is found in the secondary weapons under the Submachine guns tab, at a price of $716.800. It can be done on most of the heists but there are two I would recommend. The first is any of the bank heists from the bain contracts as long as you go loud or Hoxton Breakout from the Dentist contracts. You can do it on any difficulty but the spawn rate is better on a higher difficulty. It can be done on your own if you wanted. If you do it on your own, or with a group, it is best to have the Jack of all Trades skill. You can find it in the Technician Skill Tree and take some health kits and some ammo just in case you need it.

The Kross Vertex SMG is a secondary weapon that will coat you $716,800. Once you have the SMG I suggest setting up your skill tree with a build dedicated to SMGs so you'll be able to kill specialist enemies with it and have a lot of ammo. The gun itself is good so you won't have much of an issue with it killing anyone. The build I suggest you use is this SMG Build by LilGuyG. Just make sure you use the Kross Vertex SMG and not the ones from the video.
Once you are all setup, purchase Either a Bank Heist from Bain or White Xmas from Vlad on either Hard or Very Hard difficulty to increase spawn rate, and turn the AI off. Once you load in the heist, start killing every enemy with the Kross Vertex SMG. You will run out of ammo a lot so make sure you're picking up the ammo boxes dropped from the officers you kill. It's also the reason why I recommend bringing fully upgraded ammo bags with you. You can purchase the ammo bag asset from the heist pre-planning menu if you feel like you need it. It'll take 10 - 15 minutes until you'll be able to kill 100 enemies. The trophy will be awarded to you once the 100th kill has been made. You do not need to finish the heist. Below is a visual reference of the weapon.
