Each of the 16 missions contains three amulets, one per character. You play as each character once per mission, and will need to find their corresponding amulet while playing as them.
For Gecko and Owlette, these are virtually impossible to miss. They are directly in your path so you will always seem them. For Cat Boy, especially in later levels, there is a lot more platforming and exploration possible (relatively), so you may have to look around a little bit, but it will always be where gems are too. In addition, characters will chime in and tell you that you're near an amulet, so pay attention to this and look for the amulet after you hear this dialogue. Only a very simple and minor amount of exploration is needed to find this amulet in each mission.
As far as the achievements, the levels categories are split up throughout all 16 missions, so while you may unlock a couple achievements early, you won't unlock some, such as this one, until nearly the end of the game.
