There is actually a very simple way to get this achievement.
Build up your prison infrastructure or load up an old save until you have a capacity to hold at least 100 prisoners. Make sure that the area where the prisoners are delievered is all fenced off.
Now go into Intake. If you have built up enough infrastructure when you view the intake screen there will be 100 Minimum, 100 Medium and 100 Maximum available for a total of 300. Press X to accept them all.
My population at the time of doing this was between 50-70 with a capacity of 100
Theres going to be a lot of violent chaos so get ready. Hire as many guards as you can. When your prisoners start arriving the max security ones wont tolerate the cramped conditions very long so they will start killing each other. You just need to hold on long enough to get those 300 fortunatly, every few minutes you can keep going back into Intake and and accepting more new prisoners, these new arrivals should cancel out all of the deaths. As soon as you hit 250 the achievement will pop.
Make sure you save before hand just incase it goes wrong.
I managed to get mine to around 300. This is also a great method of getting the achievement for stopping a riot of 50. Rather than accept the full amountof prisoners just try accepting either100 Minimum or medium security, make sure you have plenty of guards ready and when the rioting begins hope press down on DPAD and bring in riot guards.
