How to unlock the Prison Hotel achievement in Prison Architect: Edition - Definitive Guide
For some reason, I can't seem to unlock this one through actually playing the game, no matter how nice I made the prisoners cells, but I found a way to accidentally unlock it.
All I did for this one is:
1. Start a new game, on Federal.
2. Build a giant holding cell, as well as a kitchen and a canteen (I'm not sure how necessary the kitchen and canteen are, but I did it anyways to be safe).
3. Build a really large deliveries area by the road.
4. Wait until your counter gets to about 125 total in the Intake area under Reports.
5. Accept all prisoners at once.
When they finally arrived, I unlocked this achievement along with Minimum Security Prison.

also did this with approx. 75 inmates.
Built around 60 comfy beds, some shower heads, windows, bookshelf, TV, radio etc. to increase the value of the cell.

I've tried this about 5 times now and have had no luck. Did you have any utilities built? How many guards did you hire? How many beds and other objects were built inside the holding cell?

Did you have to put in a reception area or a canteen or anything else?

I did not have a reception area, but did have a canteen and a kitchen. I will update the solution - thank you!

I couldn't get kashmyr7's solution to work so here's an alternative if you struggle. Start a new game on Prison Architect mode and build 50 improved or luxury cells using the quick rooms option (I did improved cells but my husband pointed out that luxury may guarantee the achievvement further). While you are waiting for the prisoner intake number to increase I built a quick rooms utilities, kitchen and canteen to make the cells fully useable and added a large delivery area for the prisoners to arrive in. I hired 10 guards to take the prisoners to the cells and then once I could take in enough prisoners I selected to intake exactly 50. It popped as the prisoners were taken to their cells.
It probably doesn't make a difference but I took 1 minimum security, 25 medium security and 24 security prisoners.

I tested luxury cells. You don't have enough cash to make 50 luxury cells. Good news is: you only need improved cells. Great guide! It works!

This achievement is bugged and doesn't seem to unlock from playing normally despite meeting the requirements.
I did it by creating a new game on County difficulty, building 51 luxury cells, a canteen, a kitchen and a yard in a fenced off area. I also increased the delivery zone size substantially however I doubt it made a difference. Finally I hooked everything up to water and power.
Once construction was 100% complete I hired a few guards and accepted 51 minimum security prisoners (in one single delivery/intake). Once all prisoners were delivered the achievement unlocked.
I hope this helps you unlock this unfortunately bugged achievement. I believe resuming a save is what breaks the achievement so make sure to start a new game and do everything without backing out to the menu.

In case anybody is still having issues with this I noticed that upon intaking a prisoner if they're caught in possession of illegal items they're put in "stasis" until they're sent to solitary, and this counts their cell as empty. To get around this I made sure to quickly hire a warden, build him an office, and then unlock prison policy through bureaucracy all before any prisoners arrived. Finally, I changed all possession of illegal items punishments to "none." When my 50 prisoners arrived I spam hired 50 guards to take them all quickly to their cells before any issues could arise and the achievement popped for me.
