This achievement is unlocked in the Mushroom Forest, which is the first area you explore after the tutorial section. Immediately after entering the area, walk to the right and interact with the red gem floating on the altar. The Queen will appear for a brief section of dialogue. Once she's done talking, the achievement should pop.

After she's done talking, you fight 3 waves of mushroom things; achievement popped for me after defating them.

I had the same experience as prakkero, the achievement did not pop until after fighting the three waves of mushroom minions.
