This is very straightforward to unlock. As it says just defeat the Tweedle Twins. So here's a few tips that should be helpful particularly on higher difficulties. The Tweedle Twins are the final boss of the Mask Mansion level. My advice is to focus on one of the twins first and take them out before going after the other. It may seem difficult to distinguish between them but there is a way to tell them apart. On their hats there's a band just above the brim. On one twin it is striped and on the other it's checkered. As for strategy this worked for me. By this point you should have your first two skills unlocked. So start of by hitting LB then RB (or vice versa). Then use the B button to quickly move in on the twin you're focusing on and spam the X button for a few hits. The twins make a slight movement just before they attack that gives you enough time to hit the B button (at least twice quickly, three to be safe) which will get you out of range of their attacks. Immediately after they land their attacks they stand around doing nothing for a second or two. So that's the ideal time to dash back in with B and spam X again. I personally found dodging/dashing to be much more useful in this battle than blocking. Keep repeating this pattern making sure to hit LB/RB when the gauge refills and you'll take them down in no time.
