This is story related and will unlock immediately after you kill the giant in the Village
farm area.
The fight itself can be quite difficult (and ammo intensive). Use the grapple to hook into the roofs and shoot the giants head until his Plaga weakspot is exposed. Then quickly focus it down whilst moving between the rooftops.
