How to unlock the Way of the Warrior achievement in Resident Evil 5 - Definitive Guide
The achievement will automatically pop up as soon as you hit the 150 enemy mark, so you don't have to wait until the end to see if you killed enough.
Tips:
- Keep the Rifle, don't give it to the CPU. On Amateur, enemies are killed pretty easily. So, if the CPU sees an enemy and kills it - they'll rob you of the kill. Get the heads up on enemies in the distance by using the rifle before the CPU sets their sights on the enemy. You might not get the Rifle in your playthrough (as it's randomized), but if you do, it makes this achievement much easier.
- Go for the "Shoot the Messenger" achievement at the same time. The Agitator Majinis take a bit of effort to lure out, so you'll rack up a few extra kills on the way. This solution on the 360 page shows how to spot the Agitators:
- You can try taking the CPU's guns away and give them a bulk of your healing items. They can heal you if you get overwhelmed and won't take any of your kills. On Amateur, damage is pretty minimal.
- Be thorough and look for the other more powerful guns available. Jill starts with a Machine Gun and Josh a Handgun. But, you can also find a shotgun, rifle, magnum and additional handgun/machine gun (the combination of guns is randomized). Very helpful, especially with the bigger Majini and Reapers. If you're thorough, you'll also find Majinis hiding in rooms or hidden areas. You can also use the large guns used for blowing up gates to take out large groups at a time.

I also suggest take your time to kill as many enemies as you can. Some areas has a lot of enemies. This achievement could also help you get Shoot the Messager achievement. There is no official counter prior to finishing it.
Among killing your one hundred fifth enemy. The achievement should pop right up. Yes you don't need to finish Desperate Escape in order to get it. I also recommend using a sniper rifle to speed up the kill counter. Please try saving your powerful weapons for the last area.
Do this on Amateur. At the onset of the game, request Josh's weapon and make sure you are the one which inflicts all damage on the Majini. If Josh helps out in any way (via melee or knife) then that particular zombie will not count towards the goal. You can guarantee defeating the 150 if done in conjunction with “Shoot the Messenger” (see below) since that trophy requires you to hang around certain areas and continually kill enemies until the Agitator Majini shows up.
As a reference, the trophy popped for me in the area right before the final rooftop battle.

Easily done on Amateur. Just take your partner's weapon away from him right at the start. (Press to bring up the item menu, then to see your partners, and choose request the weapon from him/her) Then just kill everything you see, you shouldn't run out of ammo, and it'll unlock before the end of the level. Getting this achievement is also a good way to get "The Great Escape".
