To get this achievement, you need to be playing as Moira (Press 
to switch between characters) A short while after getting the crowbar you will reach a room where you drop down into a pit and Moira makes a comment about 'bugs' and you will see a bunch of scorpion-like creatures that will come and attack you, simply kill all of them and reload your checkpoint, which will put you about 2 rooms back outside, rinse and repeat.

It's worth noting that follow-ups with Moira count as well. I got this through natural progression by making sure to use her for follow-ups. Great spot listed for farming them quick, though!

In the room that needs the rusty key will be a lot of spiders which are easily killed with the crowbar. You can kill all of them and reload the checkpoint. You will respawn in the room with the key nearby. Run back into the spider room and repeat this until you have all 50 enemy kills.
