How to unlock the Big In San Francisco achievement in Rock Band - Definitive Guide
As is the case with all "Big In..." achievements, this one requires playing in Band World Tour, so you'll need to find a (local) buddy to play with, or resort to vox (vocals & guitar by yourself).
And as well, as is always the case with the "Big In..." achievements, number of stars doesn't really matter, you only need complete each gig in every venue.
I've heard that your band's hometown can affect some of the specific gigs, but when I played San Francisco consisted of 3 venues, with gigs listed below. All but one could be played on easy, and that one other (Barroom Brawlers) required medium difficulty or better.

By far the most difficult gig is the Barroom Brawlers gig, as A) it's the longest, B) is the only gig which requires medium or better, and C) consists of a couple of the hardest songs in the game. If you have a buddy medium shouldn't pose any problems, but vox-ing by yourself can be tricky on a couple of these songs.
