Poseidon is unlocked by earning 100 stars (and possibly you have to defeat the previous bosses, Moby Dick being the one before). Then go to the top right of the map and you will find Poseidon within the triangle.
The boss fight itself is just a three lap race. Nothing much to say about it, stars give you boost, obstacles like the floor traps and turtles will slow you down. Try to avoid hitting him, because it ends up slowing you down. There are a couple of possible places to take him over, one being near the end of the lap. So if you are struggling, try to stay just behind him and take him over near the end of the last lap.
