You can activate your Power Meter once the gauge on the top right of the screen has turned white and the fretboard glows blue. To get it to this point, you need to play well avoiding missing notes and keeping your multiplier.
There is no difficulty requirement for this achievement so do it at whatever difficulty you feel most comfortable.
Activate it as soon as you get the ability to do so and then it can start building up again whilst it is in use.
An easy song to do this on is Am I Evil? as it is long enough for you to build up the power meter should you have difficulty in doing so on lower difficulties otherwise most of the first few songs for more experienced players should net you this after you have activated 3 times.
