Cumulative across levels / playthroughs. Doesn't reset when you die.
Every enemy you kill counts towards this. You'll have done 150 in the first 15 minutes or so. Just shoot away!

All of the Kill trophies will pop immediately after hitting the relevant amount mentioned in the respective description. Unless you are trying to spare every alien you possibly can, you should kill your 150th enemy before the end of the second level.
