Just watch the Blood percentage at the top of the screen. It will tell you something like 0/60, with 60 being whatever percent of all blood on screen that you have to collect to accomplish the mission. Just keep cleaning past that number until you get to 100/60.
Some general advice: First, keep in mind that several hiding places give you an option of where you exit. The truck cab outside the gas station, for example, is much more useful because you can get in on one side and out on the other. Just push the stick to the desired side before exiting. Second, remember that some of the blood pools are dynamically placed. If you just can't get one, restart the mission and see if it moves to a more advantageous spot.

Just started the game and on the very first mission that you have to clean up the blood stains I've tried twice now and it just gets up to 99 and not 100. I can't see a spec anywhere so I don't know how you are supposed to find that last 1%. Any tips?

@Mr Fiddler and OP - I would recommend The Deadly Telegraph mission as the first attempt at this. The mission in the woods kept getting stuck at 99% for me. Might be a bug but more likely it's just because it's so murky and so many objects a tiny bit of blood could be behind. The newspaper level is much brighter so it's easy to make out the blood.

Starting Contract 3, you'll find that there is a third requirement to completing the mission: evidence, ranging from 1 to 6; bodies, also ranging from 1 to 6; and blood percent, which generally runs from 30 to 95 in requirement. Thankfully, the moment you get 100%, the trophy will unlock and the level itself will retain the 100% blood completion on the Contracts menu.
