This has to be done with the big harvester you unlock on the mission "Breakfast in France" after the guy who gets you ambushed you will find the machine in his barn. Simply take this across the vast expanse of farmland and just keep driving around the area until the achievement pops as there are well over about 200 enemies here.

This is cumulative, I must have been just short in my story playthrough, as when I've gone back using chapter select it popped almost as soon as I started harvesting the first group of enemies.

Mission "Breakfast in France".
Difficulty: Any.
Follow the missions objectives and once you enter the harvester run over all types of enemies but the big bloaters and the kamikaze types and dont run over trees because they damage the vehicle.
If enemies board your vehicle you have to shoot them from the drivers seat and i recommend the assault rifle for that.
