The Meta Cannon is unlocked by completing 24 challenges in Firefight mode. This isn't difficult at all, since there are 42 missions with 2 challenges each for a total of 84 challenges. You only need to complete about 1/4 of the challenges.
Some challenges that may need additional descriptions:
Kills in flip/while upside-down: while in the air in slow-mo, look up or down to "flip" your character so that her feet are not facing the floor. "In flip" doesn't require you to be quite as flipped as "while upside-down" does.
Explosion kills: Kill an enemy with an explosive barrel. You can do this by simply shooting the barrel, but it's easier to pick up the barrel with the Gravity Gun arm cannon and throw it at an enemy.
Once you've completed 24 challenges and unlocked the Meta Cannon, go to Mission Select and complete the City level with it. Note that using mutators may prevent the achievement from unlocking.
