The "Slayer's Gauntlet" Challenge Tomb is the last part of "The Serpent's Heart" DLC. To access it you'll play through the new mission. The entirety is shown in my Walkthrough below with a run time of 41 minutes. The specific tomb challenge is at 22:12 in the video.

I would like to beat the living shit out of whatever dev thought riding a raft in a circle, while trying to dodge multiple traps and fire while trying to shoot a target so small on my tv its next to impossible unless standing still. I downloaded this day it came out and try about 2-3 times a day then quit before I break something

Using the shotgun on the targets to stop the fire makes things much easier
