To earn this achievement you *CAN NOT* purchase Will or Darkness from Missy (green lady) in the tower.
Missy will tell you what stage you missed any Will or Darkness chests in, and offer to sell you them at 4,000 gold each. DO NOT buy them from her, or you will miss the achievement and have to unlock it on a separate playthrough. Do, however, use this information to see which levels you have missed chests in.
If you have collected all the Will and Darkness chests from the levels you have played so far, when speaking with Missy her last sentence will be "I've got nothing to sell you. You're so meticulous, Specty!"

Everything evllmatt says is correct and useful, do not buy any of the missed wisps from Missy, but you can use her to find out which stages you are missing wisps from.
Most of the wisps are easy enough to find. Some are already in plain sight (Plains, King Knight, etc), but if you've hunted for secret areas in Shovel Knight before you'll have no trouble finding all of the wisps except maybe for one -- Plague Knight's.
His is tricky, because the entrance to the secret area isn't as obvious as the usual one-tile-doesn't-match on the wall -- it's actually above you. Below is a screen shot of where you can find the entrance on Plague Knight's stage. It's in the room with a checkpoint and two potion dispensers. If you hit a potion from the the rightmost dispenser back up, it will reveal the ladder to the wisp.
