I just unlocked this and I can confirm that it also includes hostile animals. Killed a wolf and it unlocked.
So far, it seems like you should just replay "The gates to Hell," as it's a quick succession of enemies in a row. Other missions that can be quickly completed and require you to kill several demons are also very helpful.
Give me some feedback if I'm incomplete on this answer. This game was just released.

Just to add to this, the achievement for 1,000 kills popped for me when I killed a non-hostile buffalo. So looks like non-hostile animals count towards this achievement.
