This is story related during the sixth campaign mission, Tiergarten Flak Tower and is therefore unmissable.
Proceed through the level, up and over the flak tower, then down it again until you reach the winch room. If you make it across without being seen you will earn this achievement as well:
The Kilroy was Here achievement in Sniper Elite V2 worth 50 pointsMake it through the tower to the winch room without being spotted
Next you will see a cut scene and then you need to shoot Müller. He may not be visible initially but if you shoot a few of his guards he will come into view. When making the shot on him remember to press RB for focus mode as it does really help in making a shot of this distance.
Using focus mode also goes towards helping you get:

This trophy is story-related and cannot be missed via normal campaign progression.
You will unlock this trophy for completing Chapter 6: Tiergarten Flak Tower.
First you will need to make it to the base of this huge tower and then you must ascend it. Outside of the tower, the Germans are fighting the Russians. Use this to your advantage! You want to focus on shooting more of the German troops as there is a limited supply of Russian troops.
Once at the bottom of the tower, try to quietly kill and make your way to the top. When you reach the top you have to go across the roof to the other side and go to the winch room to have an opportunity to take down your target. After the target is down, you must escape the tower.
The trophy will unlock at the end-level screen.

This will unlock mid-mission. When you reach the top of the tower, you will snipe Muller through a hole in the wall.
