After the prologue and upon starting Act 1, you will have access to the open world map. On it, you will notice "?" scattered around. Traveling to these points and looting containers, rescuing civilians, analyzing tracks, etc. in that area will count as completing the Point of Interest. You will also get an on screen prompt that you have completed one.
